Introduction
Several evidences have shown that limiting caloric intake is an effective non-pharmacological strategy to reduce cardiometabolic risk factors and to improve heart function thereby delaying the onset of cardiac diseases. Our preliminary data shows that caloric restriction (50% of food intake) reduced vascular contraction and improved vasodilation while reducing systemic inflammatory status and hypertrophy markers, while increasing cardioprotective atrial natriuretic peptide. This research will prove whether these data are related to a possible prevention of cardiovascular damage related to inflammation involving endothelial dysfunction, cardiac hypertrophy and neointima formation.
Methods
Mice will be kept on 50% caloric restriction starting at birth up to 30 days after the wean. Neointima formation will be used in a surgical model for carotid artery and cardiac hypertrophy; while pharmacologically, mice will be treated with isoproterenol. Heart and aortic function will be evaluated by Langerdoff organ chamber experiment. Morphological changes will be evaluated by light and confocal microscopy. Protein expression will be quantified by Western blot and immunostaining.
Results
The research will shed some light on the reasons why caloric restriction presents cardiovascular protective effects even under pathological stimuli such as changes in blood flow or hyper-stimulation of beta-adrenergic receptors and elucidate the mechanisms advancing the effect.
Conclusion
By our hypothesis, caloric restriction upregulates nitric oxide pathway reducing smooth muscle and cardiac cells hypertrophy and inflammatory genes.
