Metabolomic characterization of Aspergillus flavus strains by UHPLCHRMS and molecular networking: chemical diversity and bioactive potential.

  • Aspergillus flavus, belonging to the Flavi section of the Aspergillus genus, is a species of filamentous fungus widely distributed in agricultural and postharvest environments and is recognized for its ability to produce aflatoxins. In this study, Aspergillus flavus strains isolated from soybean plants cultivated in southwestern Piauí were characterized by ultrahigh-performance liquid chromatography coupled with high-resolution mass spectrometry UHPLCHRMS combined with molecular networking. For this purpose, methanolic extracts from fungal isolates FS29, FS30, FS42, RS11, RS32, and RG8 were prepared and analysed by UHPLCHRMS in positive and negative ion modes. The processed data were compared with spectral libraries, and the molecular signatures were identified according to the elution order, MS/MS fragmentation pattern, molecular formula, and database search. Additionally, spectral data from A. flavus strains were used to generate molecular networking via the feature-based molecular networking FBMN method on the Global Natural Products Social Molecular Networking GNPS platform. A total of 42 metabolites, including peptides, organic acids, alkaloids, trichothecenes, nitro compounds, terpenes, phospholipids, and phenolic compounds with antimicrobial, phytotoxic, and plant growth-promoting activities, were annotated in A. flavus methanolic extracts. Citric acid and ferulic acid are associated with antimicrobial activity. Ravenic acid and phomalenic acid A showed antibacterial potential, while alkaloids such as ditryptophenalin and cyclopiazonic acid demonstrated cytotoxic and antimicrobial activities. Related mycotoxins, including patulin, NX2 toxin, and diacetoxyscirpenol, which are associated with the inhibition of protein synthesis and fungal virulence, were also detected.

O evento promoveu a integração entre academia e setor produtivo por meio de conferências, mesas-redondas, minicursos e apresentações de trabalhos científicos, incentivando a troca de conhecimentos e experiências sobre temas como microbiomas do solo, bioinsumos, controle biológico, abordagens ômicas e tecnologias inovadoras para o agronegócio. Além disso, ofereceu atividades de formação complementar com enfoque prático, contribuindo para a capacitação técnica e científica dos participantes.
