Different Doses Of Curcumin Exert Antioxidant Properties, Decreases MMP-2 Activation And Reduces Vascular Remodeling In L-name Induced Hypertension

    Introduction: Increased reactive oxygen species (ROS) formation is involved with matrix metaloproteinases (MMPs) activation and vascular remodeling. The antioxidant effects of Curcumin are widely known but is still uncertain if curcumin decreases vascular remodeling and MMPs in hypertension. Despite curcumin gained considerable attention for its multiple pharmacological activities; its bioavailability is low. The aim of the present study was to evaluate if different doses of curcumin decreases vascular remodeling and MMP-2 activity in hypertensive rats.

    Methods: Male Wistar rats were treated with L-name or water. Both groups were also treated with two different doses of curcumin (50 (CUR50) or 100 mg/Kg/day (CUR100)) or carboxymethilcelulose (vehicle for curcumin). Blood pressure was evaluated by plethysmography. ROS formation and MMP-2 activity were evaluated by reaction with dihydroethidium (DHE), thiobarbituric acid (TBARS) and zymography. The cross-sectional area (AST) and medium/lumen (M/L) were evaluated in the aorta. The statistical comparisons were assessed by 2-way analysis of variance or by t-Test. A probability value <0.05 was considered significant. (Approved ethics committee number: 13/2017). Results: Blood pressure (BP; 176±4mmHg) and TBARS (77±7mM) were higher in the L-Name group when compared with control (125±3mmHg and 62±4mM; p<0.05). Curcumin treatments decreased BP (p<0.05) and TBARS in hypertensive rats independent of the dose (CUR50:55±3mM and CUR100:63±3mM, p<0.05). DHE assay confirmed the antioxidant properties of Curcumin, such as observed by TBARS. MMP-2 activity was 46% higher in the hypertensive group (p<0.05) and was reversed by curcumin treatment independent of the dose (p<0.05 vs L-name). Higher levels of AST and ratio M/L was found increased in hypertensive rats (p<0.05 vs control group) and were attenuated by curcumin, being significantly different in the dose of 50 mg/kg/day (p<0.05 vs L-name). Conclusion: The antioxidant properties of curcumin may be associated with MMP-2 activity reduction and improvement in vascular remodeling during hypertension.
