Introduction: Diffuse cutaneous leishmaniasis (LCD) is a disease that can be caused by the genus Leishmania amazonensis. It is a pathology associated with skin and mucous membranes, but morphological changes in vascular tissue have shown that this disease can cause serious changes in the vasculature. However, the possibility of vascular function being affected is unknown.Thus, the aim of this study was to evaluate the impact of Leishmania amazonensis infection progression on vascular reactivity.
Methodology: Male BALB/c mice aged 8-12 weeks (25-30g) were used, divided into groups: control and infected. The infection occurred by subcutaneous inoculation of the hind paw with a parasitic load of 105. The animals were killed between 30 and 60 days. Afterwards, the vascular reactivity process was performed to obtain the vascular contraction and relaxation responses.The statistics were determined by the Student t test or One-Way ANOVA test and Tukey's post-test. Approved by the Ethics Committee (CEUA UFMT- 23108,920630/2017-56).
Results: LCD infection caused an increase in vasoconstriction within 30 days (106,4±5,02 vs. 131,2±5,50) and 60 days of infection (106,4±5,02 vs. 141,9±4,80), with greater dysfunction associated with disease progression. There was a reduction in the percentage of relaxation in the arteries previously incubated with L-name (nitric oxide synthase endothelial inhibitor), within 30 days (30,37±0,34 vs. 21,86±0,39) and 60 days of infection (30,37±0,34 vs. 25,59±0,65), compared to the control group.
Conclusion: Analyzes of vascular reactivity showed that infection with Leishmania amazonensis caused vascular changes in vasoconstriction and vascular relaxation, according to the time of progression of the infection.
