In addition to endothelial dysfunction, heart failure (HF) promotes the loss of the anticontractile effect and whitening of PVAT of the thoracic aorta. Recommendations suggest that in addition to the practice of aerobic exercises, resistance exercise should be added to training programs and promote benefits to the cardiovascular system. Objective: To evaluate the effect of combined training(CT) on the functional and phenotypic adjustments in the PVAT of the thoracic aorta of rats with HF. Methods: CEUA 053/2014. Male, Wistar rats were submitted to occlusion of the coronary artery(HF) or fictitious surgery (SO). After 4 weeks, the rats were divided into: untrained (uSO:n=36 and uHF:n=38) and resistance (ladder) and aerobic (treadmill) training, on alternate days (5x/week/8 weeks, tSO:n=26 and tHF:n=21). At the end of training, thoracic aorta rings with endothelium (E+)in the presence (+)or absence (-)of PVAT were mounted on a wire myograph and concentration-response curves to phenylephrine(PHE) were performed. Gene expression analysis for white, beige and brown adipose tissue markers, as well as inflammatory markers were performed in PVAT. ANOVA (p<0.05*vs. uSO and #vs. uHF). Results: PHE-induced contraction was increased in rings of uHF compared to uSO and CT was able to partially reverse this effect only in the presence of PVAT (Emax of E+/PVAT+ rings:uSO:3.6±0.3, uHF:8.6±0.4*, tHF:6.6±0.4mN/mm*#). PVAT of uHF present increased gene expression of the white adipose tissue (TCF-21) and reduced of beige and brown adipose tissue markers (EPSTI-1 and PRDM-16, respectively); CT reversed these changes. In addition, CT reduced the higher pro-inflammatory markers (TNF-alpha,IL-6 and MCP-1) and increased the lower gene expression of IL-10 (anti-inflammatory marker) observed in PVAT of uHF. Conclusion: CT repairs the PVAT whitening phenotype, pro-inflammatory status and the dysfunction observed in thoracic aorta of HF rats. These effects together suggest that CT is a beneficial milieu to ameliorates the vascular dysfunction in HF.
