  Preeclampsia (PE) is a multifactorial and multi systemic disease that reaches great part of pregnant women and is responsible for a high rate of maternal and fetal morbidity and mortality. These cases are emphasized in poor and in development countries, like Brazil. Despite being one of the most severe disturbs among the ones that occurs during the pregnancy, little is known about PE etiology. There is the hypothesis that pseudo-vasculogenesis, a limitation on the invasion of spiral arteries by the trophoblast, is responsible for generating placental ischemia, that causes liberation of placental factors in the mother’s bloodstream and, consequently, to the dysfunction of vascular endothelium. The reduced bioavailability of Nitric Oxide (NO) and the oxidative stress caused by the imbalance of antioxidant agents and oxidants in the organism contribute to such endothelial dysfunction. Antioxidant effects of phytochemicals found in foods and drinks has proven to be efficient and the flavonoids, polyphenol phytochemicals, have shown a reduction in blood pressure, as indicate studies. Quercetin, a flavonoid found in grape juice, can activate the endothelial cell nitric oxide synthase (eNOS) via a protein phosphorylation cascade. The eNOS is responsible for producing NO, so it’s activity, boosted by Quercetin, ends up causing vasodilation and, consequently, decrease arterial pressure. Observing these effects in hypertensive diseases, in vitro tests will be done and human umbilical vein endothelial cells (HUVECs) will be incubated with preeclampsia pregnant women plasma, hypertensive and healthy, in presence and absence of Quercetin for the effects evaluation of this phytochemical in PE, verifying: eNOS expression activity, nitrite quantification to evaluate NO formation, intracellular NO, endothelial dysfunction markers, cell viability and reactive oxygen species quantification. This study aim to bring important information about the use of Quercetin in prevention and treatment of PE.
