Obesity in children and adolescents has increased considerably in recent years and it is often associated with risk factors for cardiovascular disease. These comorbidities, especially hypertension, are a consequence of the altered production of adiponectin by the adipose tissue. Genetic variants at the ADIPOQ locus have been associated with adiponectin concentrations and cardio-metabolic complications. However, no studies have examined these associations in Brazilian children. We examined the effects of ADIPOQ SNPs on adiponectin and its high molecular weight oligomer (HMW) levels. We recruited 299 children and adolescents (58.2% women, age 11.2 ± 3.1) divided into: control (CT; N=143); obese (OB; N=106) and obese hypertensive (OBH; N=50). All participants were interviewed for data collection and underwent anthropometric measurements, genetic and biochemical analysis. Plasma adiponectin concentrations were determined in EDTA plasma by ELISA and rs266729 and rs1501299 genotypes were determined by Taqman Allele discrimination assays. Unpaired t-test, Mann Whitney test, and Kruskal-Wallis tests were used when appropriate to compare groups; P<0.05 was considered statistically significant. OB subjects carrying CG+GG genotypes for rs266729 showed lower HMW adiponectin and HMW/adiponectin ratio when compared to individuals with genotype CC (CC: 2.7±1.4 vs. CG+GG: 2.0±1.2; *p<0.05; unit: µg/mL; ratio). We have not observed the influence of genotypes on adiponectin levels in the CT and OBH subjects (P>0.05). SNP rs1501299 showed no differences between genotypes. These results suggest that genetic variants at the ADIPOQ (rs266729) is associated with lower HMW and HMW/adiponectin ratio in Brazilian obese children and adolescents.
