Evaluation of ?-3 Enrichment on the Vascular Changes in Rats Offspring Subjected to Maternal High-Fat Diet During Pregnancy and Lactation: Focus on the ROS/ADAM17/ACE2 Pathway

  • The consumption of a high-fat diet is a risk factor for many comorbidities during the human development such as metabolic disorders and phenotypic changes, especially It has a direct influence in the progress of cardiovascular diseases in adulthood. These changes may be associated with an imbalance between the antioxidant defense system and the production of reactive oxygen species (ROS). Angiotensin-II (Ang-II), the main vasoconstrictor of the renin-angiotensin system (RAS), plays a central role in oxidative stress, inducing an increase in the production of ROS and endothelial dysfunction. However, the diet rich in omega 3 (?-3) induces the production of anti-inflammatory substances and cardiovascular protection. The aim of this project is to investigate the effects promoted by ?-3 enrichment on the vascular changes of rats’ offspring subjected to maternal high-fat diet during pregnancy and lactation, focusing on the ROS/ADAM17/ACE2 pathway changes. Primiparous albino rats from the Wistar strain (Rattus norvegicus), weighing between 220 and 260 g, will be used, and the determination of pregnancy will be carried out. From the first day of gestation until the 21st day of lactation the progenitor rats will be placed in individual cages with their respective diets with ad libitum feed and water. In order to assess the influence of ?-3 enrichment on vascular function, experiments will be performed to better verify the vascular reactivity in mesenteric artery of rats. To analyze the oxidative stress in the aorta and blood of different groups of rats' offspring, lipid peroxidation will be measured by TBARS assay. In addition, the expression of ADAM17 and ACE2 and evaluation of mRNA expression of proinflammatory cytokines (IL-6, TNF-? and CX3CL1) in the aorta of rats will be performed accordingly. The project will be submitted to the ethics committee on animal use (CEUA/UFPE) and the experiments will start after approval.
