Evaluation of Cardiovascular Repercussions in First Generation Adult Descending Rats in an Experimental Model of Gestational Hypertension

  • Preeclampsia (PE) is a disease of unknown etiology that occurs during pregnancy. There are not enough studies to describe the cardiovascular repercussions in the offspring of mothers with PE. The objective of this study was to evaluate the repercussion on the cardiovascular system of the offspring. Twelve female (Wistar) rats were obtained, between 12 and 16 weeks of age, weighing between 200 and 250 grams and divided into 2 groups, being group 1 control (N6) and group 2 hypertensive (N6). CEUA 1206. To induce hypertension, we injected the rats intraperitoneally on day 1 of gestation with 12.5 mg of deoxycorticosterone acetate, followed by day 7 and 14 of gestation of 6.25 mg and their drinking water was replaced by a solution NaCl (0.9%). After birth, the offspring, 20 males, descendants of group 1 (N10) and group 2 (N10), were weighed on days 1, 10, 21, 60 and 90 of life, and the pressure was measured on days 60 and 90 of life. On day 90 of life, the offspring were euthanized, vascular reactivity was performed on abdominal aortic rings (with intact and mechanically removed endothelium), submitted to a concentration-response curve to KCl, phenylephrine, acetylcholine and sodium nitroprusside. The offspring of hypertensive mothers had 26% less birth weight. Blood pressure in adulthood was considered normal. In vascular reactivity, no statistical differences were found in any of the curves. When it comes to partial results, we can conclude that the offspring of hypertensive mothers have decreased birth weight. However, the organism can use compensatory mechanisms during the growth of the offspring. We believe that the same reasoning can apply to that observed in blood pressure parameters. In addition, in general, it was observed that the rings tended to be more sensitive to the drugs in the study. The error considered is P<0.5.
