Bothrops Moojeni Type 1 Metalloprotease Reduce Vascular Remodeling In A Model Of Renovascular Hypertension

    It has been shown that type 1 metalloprotease from Bothrops moojeni cleaves angiotensin I in vitro, decreasing the levels of angiotensin II (Ang II). The renovascular hypertension model of two Kidney one clip (2K1C) causes high blood pressure by improving Angio II and is widely used to assess drugs' effect. This study hypothesis that metalloproteinase from B. moojeni decreases blood pressure and aorta remodeling in vivo. Therefore, this study aimed to evaluate the effect of B. Moojeni type 1 metalloproteinase (Bm1) on aortic remodeling in 2K1C rats. We used male Wistar rats with 180-200g (CEUA nº 1693010319). Sham-operated or hypertensive rats were treated with vehicle, Bm1 (metalloproteinase PI BmooMPalpha-I of B. Moojeni 1?g/kg), Losartan (10mg/kg), or a combination for four weeks. Systolic blood pressure (SBP) was measured weekly by tail-cuff plethysmography. Aortic morphometry, elastin, and collagen contents were evaluated using hematoxylin/eosin, orcein, and Picro Sirius red-stained sections. Treatment with Bm1, Losartan, or the combination of drugs attenuated 2K1C induced hypertension, decreasing the blood pressure in 30 mmHg. No difference was observed between the treatments (p>0,05). Increased cross-sectional area (CSA), elastin, and collagen contents were observed in the aorta of animals 2K1C compared to the sham group (p<0,05). Treatment with Bm1, Losartan, or the combination decreased aortic hypertrophy and elastin contents in aortas of 2K1C rats compared to the sham group (p<0,05). In collagen contents, just the treatment with Bm1 and Losartan decreased the collagen in aortas of 2K1C compared to the sham group (p<0,05). In conclusion, treatment with Bm1 attenuate hypertension by lowering blood pressure, elastin, and collagen contents in the aorta, decreasing hypertrophy. Therefore, Bm1 has potential as an antihypertensive, but further studies are needed to elucidate the mechanism of action.
