Elevated levels of matrix metalloproteinase 2 (MMP-2) play a role in the pathophysiology of cardiovascular diseases. Patients with periodontal disease secrete more MMP-2 into the plasma, which is associated with coronary heart disease progression and atherosclerosis. MMP-2 knockout mice exhibit reduced atheromatous plaques lesions. This study hypothesis that MMP-2 changes blood cells and plasma lipids levels. This study aims to evaluate the effect of MMP-2 on the hematological and biochemical profile of mice after four weeks of treatment with recombinant MMP-2. Methods: MMP-2 was expressed and purified in E. coli bacteria with pET5a_rhMMP-2 clone. Quantification of MMP-2 was performed using a BCA kit. MMP-2 activity and purity assessments were determined by gel zymography and silver staining. C57BL/6[Wild type (Wt)] mice, n° 6175230518 CEUA, with 7 weeks, 25g, were separated into two groups. MMP-2 group was treated with rhMMP-2 (150 ng/g of animal), and the control group was treated with saline (0.9% NaCl). Afterward, the samples were analyzed by automated count and by biochemical test kits (Labtest). Results: There was no difference in red and white blood cell counting between the control and MMP-2 groups (P>0.05). Renal function parameters were not affected either, except for urea serum level analysis, between the control (81 ± 7 mg/dL, n=7) and MMP-2 group (42 ± 4 mg/dL, n=7, P<0.05). Regarding the lipid parameters, animals treated with MMP-2 showed an increase in cholesterol and a decrease in triglycerides plasma levels, compared to the control group (Control cholesterol: 47 mg/dL ± 3, n=7 vs. MMP-2: 63 ± 5 mg/dL, n=7, P<0.05; Control triglycerides: 69 ± 13 mg/dL, n=7 vs. MMP-2: 25 mg/dL ± 5, n=7, P<0.05). Conclusion: An increase in MMP-2 levels can play a role in modulating cholesterol and triglycerides metabolism. New studies will be conducted to assess how MMP-2 alters the plasma lipids.
