Introduction: Peryllic Alcohol (PA) is a monoterpenoid present in the composition of the essential oil of many vegetables, being the majority constituent of mint, mint and ginger oils. It is widely used in the cosmetics, perfumes, cleaning products and food industries due to its pleasant aroma. Some pharmacological activities of PA have already been reported, such as: antifungal, anticancer and vasorelaxant in rodent arteries. However, despite its potential vasorelaxant effect, it has not yet been described in human umbilical veins. Therefore, the present study is unprecedented in this model. Objective: We sought to evaluate the effect of PA on the smooth muscles of the human umbilical vein (HUV). Methodology: Sample collection and processing were approved by the Human Research Ethics Committee of the Universidade Regional do Cariri – URCA (nº 3,832,881) and by the Ethics Committee of the São Francisco de Assis Hospital and Maternity, both in Crato, Ceará, Brazil. The HUVs were isolated, cleaned and sectioned into 3-4mm rings. The HUV rings were suspended in an organ bath, with a capacity of 10mL, in a Krebs Henseleit solution with physiological conditions, under a carbogenic mixture, and a pH of 7.4 at a temperature of 37 C°. Results: To evaluate the effect of PA on the basal tone of HUV, increasing and cumulative concentrations of PA(1-1000 µM) were added to the preparations. The PA changed from the HUV basal tone from the concentration of 3 µM (p<0.05). PA (1-1000µM) was able to completely relax the contractions induced by 60mM KCL (electromechanical coupling) and the contractions induced by 10 µM serotonin (pharmacomechanical coupling) in HUV, being statistically significant from the concentration of 100 µM for both preparations, with EC values of: 161.1 ± 6.9 µM and 269.8 ± 7.8 µM respectively. The PA (1-1000 µM) was also able to relax the contractions evoked by HIST 10 µM (pharmacomechanical coupling) with maximum effectiveness, obtaining an EC50 of 123.2 ± 7.1 µM, and in this route the PA showed greater potency pharmacological when compared to EC50. Conclusion: The data found demonstrate that PA has a vasorelaxant effect on HUV with greater pharmacological potential in the histaminergic pathway. These data demonstrate new possibilities in the treatment of vascular dysfunctions of the umbilical vessels, using natural products, such as perillyl alcohol.
