INTRODUCTION: Cnidoscolus quercifolius Pohl (Euphorbiaceae) is an endemic plant of the caatinga, with ethnobiological and ethnopharmacological reports for its importance for the development of the semi-arid region and for being widely used in traditional medicine [1]. Records in the literature indicate the presence of phenols such as quercitin and eugenol [2]. AIMS: To analyze the total phenolic content of the ethyl acetate extract of C. quercifolius peels and evaluate the antioxidant activity using different methods. METHODS: Ethyl acetate extract from C. quercifolius peels (EAECq) (1.5 kg) was obtained using the Soxhlet extraction system with ethyl acetate and concentrated in a rotary evaporator, obtaining a yield of 4.35%. Quantification of total phenols resistant to the Folin-Ciocalteu oxidation method, using gallic acid as standard, and results expressed in mg equivalent of gallic acid per g of extract (mg GA/g Ext.). Antioxidant activity was evaluated using the DPPH• free radical scavenging and ABTS+ free radical scavenging assays, both at different concentrations (ranging from 5.0 to 1000 µg/mL). The tests were read using an Elisa and UV-visible spectrophotometer at 518 nm and 734 nm, respectively, using ascorbic acid as the standard for both tests. RESULTS AND DISCUSSION: In the quantification of total phenols, the presence of 0.043mg GA/g Ext was verified. For antioxidant activity, EAECq did not present significant values ??in the elimination of the DPPH• radical, making it impossible to calculate the IC50 at the concentrations tested. The same happened in the ABTS+ method, but as expected, ascorbic acid was effective at all concentrations. CONCLUSION: Therefore, the ethanolic extract of C. quercifolius peels contains a significant amount of phenols and has low antioxidant potential compared to the DPPH• and ABTS+ methods. Additional research is needed on antioxidant action through different reaction mechanisms.
