QUANTIFICATION OF TOTAL PHENOLS AND ANTIBACTERIAL ACTIVITY OF ETHANOLIC EXTRACT FROM THE SHELDS OF Ziziphus joazeiro Mart.

  • INTRODUCTION: Ziziphus joazeiro Mart (Rhamnaceae), popularly known as "juá", “joazeiro” is an endemic species of caatinga and has high commercial value. In traditional medicine it is used as; oral antiseptic, anti-inflammatory and bactericidal. AIMS: Quantify the total polyphenol content of the ethanolic extract of Z. joazeiro bark and evaluate the antibacterial and antibiotic modulating activity [1]. METHODS: The peels (484 g) were collected in Exu, Pernambuco, Brazil and subjected to cold extraction with ethanol for 72 h. Then the solvent was distilled in a rotary evaporator to obtain the crude extract (yield 4.71%). The quantification of total phenols followed the Folin-Ciocalteu oxidation method at concentrations ranging from 0.05 to 5.0 mg/mL, readings were taken at 760 nm on a UV-vis spectrophotometer [2]. The analysis was carried out in triplicate and the content of phenolic compounds was calculated from the calibration curve using gallic acid. The antibacterial activity was carried out against Staphylococcus aureus (ATCC 10) and Klebsiella pseudomonas (KPC) following the microdilution method in BHI with the extract diluted in distilled water at concentrations of 512, 256, 28, 64, 32, 16 and 8 ?g/ ml. The modulating activity of antibiotics was carried out with the bacterial strain that showed activity at the concentrations tested with the antibiotics benzylpenicillin and amikacin. Both activities were read using the colorimetric method by adding 25 ?L of resazurin. RESULTS AND DISCUSSION: When quantifying total phenols, the presence of 45.795mg GA/g was verified. Antimicrobial assays demonstrated that the Minimum Inhibitory Concentration (MIC) was 512 ?g/mL, against the bacteria Klebsiella pseudomonas and Staphylococcus aureus. For the modulating analysis, the extract, when associated with benzylpenicillin, demonstrated synergism and for amikacin, it showed antagonism against Staphylococcus aureus, while for Klebsiella pseudomas, the results were not significant. CONCLUSION: The extract presents several secondary metabolites such as phenolic compounds and flavonoids, providing biological activities that can contribute to the development of therapeutic resources against bacterial infectious processes. ACKNOWLEDGEMENT: CAPES and CNPq.
